Take home exam question for Glycosylation section. Hand in by Monday morning, Jan 17, 2005 Look up Sda antigen. Googled it if you like. Get to know the epitope. I do not have the “standard” answer to some of the following questions. All you need is to convince me that your answers are supported by cited references. You don’t have time to read through all the papers and references and you are not writimf a dissertation or thesis for that. Just give short and direct answers to demonstrate that learnt your course well and you know some of the basics of glycosylation. 1. What’s the precise oligosaccharide structure constituting this epitope? Give both text and figure representation, indicating clearly the stereochemistry, anomericity and linkage. 2. Give 2 example of glycoprotein (with reference) where this epitope has been identified and characterized. What evidence are there for its identification? On which glycan chain carrier was it found? Can you have more than one Sda epitope on one single glycan chain or one single glycoprotein? 3. Any evidence that is also found in glycosphingolipid? If so, on which structural series? 4. Is there am existing and/or commercially available monoclonal antibody for its detection? 5. Is there any publishing data indicating its regulated expression in different tissues, at different developmental or disease stages? Is it occurring widely in all mammals? 6. When evidence, if any, do you think has clearly demonstrated its biological importance? 7. What is known about its biosynthesis – which glycosyltransferases are needed? Which is the one required specifically to generate this epitope? Has it been cloned and characterized? Was it in the glycosyltransferase databank? 8. Is there any evidence for its binding partner, i.e. a lection or receptor that may recognize specifically this epitope? 9. If you identify a similar epitpoe on your protein and would like to study if further for its biological implication or not for the functioning of your protein? 10. For an in vitro binding assay to look for putative receptors, you nay need to purify the protein of interest carrying this epitope. Which expression system can you use, why? 11. After all these hard work trying to meet the deadline, do you think Sda is and interesting or boring epitope of no consequence? Where dose its name come from?
