Monoclonal antibody is the first generation of HIV vaccine, and it has failed so far, and it is expected to fail in the foreseen future. The primary challenge of using monoclonal antibody against HIV is two folds. First antibody can only target the coating proteins of free virus, which limits the number of proteins and sites antibody can target. Secondly, the two env proteins, gp41 and gp120, are highly variable such that HIV can easily mutate at antibody binding site (epitopes) and thus escape monoclonal antibody targeting.
