有幾個跟Cy5有關的問題 希望知道的人能夠給點指點^^
Red laser light that produces fluorescent emission from Cy5 can also
switch the dye to a stable dark state. Exposure to green laser light
converts Cy5 back to the fluorescent state, but the recovery rate
depends critically on the close proximity of a secondary dye, Cy3.
主要是有關這一段的問題.在看到這篇paper以前我認知中的Cy5是照紅光就會亮,
然後就一直亮到它photobleach為止.所以我看不懂為什麼red laser會
"switch the dye to a stable dark state".而且這裡green laser的存在目的是什麼呢?
如果不照green laser, 一直照red laser, Cy5就不會再亮了嗎?
"green laser light converts Cy5 back to the fluorescent state",
fluorescent state不是指excited state嗎?可是在paper裡面照綠光的
時候是沒有emission的. 這幾部分我真的很疑惑.
另外請問Cy3跟Cy5之間的距離會影響recovery state的問題,這應該從哪個角度去解釋呢?
因為我查相關資料,裡面也只是提到這個現象,但並沒有詳細解釋為什麼?
