大家好 想請教兩個問題..
就是我已經有一段candidate promoter region(element)
它會activate(or repress)我的target gene (reporter assay)
接這就要找出是否有transcription activator (or repressor)
會bind到這段element
1.請問有哪些方法可以找呢?
我有想到One hybrid ,因為有library,所以可藉由reporter activaity來釣出
會bind的protein而且是有activate (or repress) 的作用
2 再paper上有看到抽crude protein extract然後去跑EMSA
然後再用chromotography把這個protein純化出來 ?? 這原理是?)
