[求救] 293T cells & starvation

不知道板上有人試過：
starve 293T 24 hours after transfection.
stimulate with 10% FBS for 10 mins

目前看到的 papers 都是用 HEK293 with FBS
或是用 293T but with TPA or EGF stimulation

或用 NIH3T3 with FBS
(but Ca phosphate transfection efficiency is poor with this cell line)

我想要大量增加 ERK activation/phosphorylation
但是看過幾篇 papers 後還是不知道 EGF, TPA 和 FBS
在 293T 上的效果/差別在哪，有人有建議或經驗嗎？

→ ranilla:對細胞而言，293T比293多一個 SV40 large T antigen
→ ranilla:對藥物而言，只是使用不同的方法來activate ERK
→ ranilla:來確定p-ERK的功能，去除這些藥的side-effect
→ ranilla:而最後的結果，則需overexpress ERK 上游，knock down下游
