標題[求救] nickel column

小弟是化學系出生的
目前在試作nickel column gel的合成
確定可以把nickel接上去了
卡在不知道如何定protein的loading量
主要問題如下：
沖提是用0.5 M imidazole
但是發現imidazole也會和Coomassie Brilliant Blue G-250作用
呈現藍色和protein一樣
這樣是只能作透析嗎 還是有其他方法?
另外有看到可以用低pH值的buffer降低nickel和protein的作用
將protein沖出
有人這樣用過嗎?
