國家理論科學研究中心學術演講 2009 NCTS Seminar on Statistics Speaker: Professor Kuo-Bin Li 李國彬 (National Yang-Ming University) Time: Friday, Oct. 23, 2009 (1)11:30 am-1:00pm, group meeting (2)1:00-2:30pm, seminar Topic: Applying Computational Sequence Analysis to MicroRNA-mediated Gene Regulation Abstract: MicroRNAs are small noncoding RNAs that are predicted to regulate the expression of approximately one-third of all human genes. This regulation occurs post-transcriptionally through miRNA binding to mRNA targets leading to subsequent target degradation or inhibition of translation. I will focus on three bioinformatics applications to miRNA-mediated gene regulation. In the first application, the dysregulation of miR-224 was shown to be related to hepatocellular carcinoma. To identify the putative target genes of miR-224, we developed a computational filtering system that aims to reduce the number of false positive predictions. Experimental biologists often select convincing putative target genes based on the observation of negative correlation in miRNA/mRNA expression profiles. To investigate the effectiveness of this strategy, we also conducted a systematic survey on the correlation between miRNA and mRNA expression profiles using data generated from NCI-60 cancer lines.
