Verax weighted microcarriers C. Biomat Informatrix porous microcarriers Processes used to make these microcarriers: A. Percell porous gelatin These microcarriers were made from an 8% gelatin solution by a double emulsion technique: surfactant was controlled by a double emulsion technique: surfactant was added to a warm gelatin solution and the solution was saturated with droplets of toluene containing additional surfactant. These toluene droplets formed a stable emulsion that created internal voids in the microcarrier; the pore size was controlled by the vigor of mixing of the solution. Addition of excess toluene created droplets of this emulsion in the toluene. The gelatin was then chilled, washed with ethanol and dried. The gelatin particles were rehydrated in phosphate buffer, crosslinked with glutaraldehyde, washed, autoclaved, and re-dried. In the resulting product, the voids were interconnected and thus accessible to cell ingrowth. (Nilsson et al.) B. Verax weighted microcarriers The particles are made from dry milled bovine native collagen, ideally from tendon, using a freeze-drying process. The collagen is suspended at about 0.5-1.5%(w/w) in dilute acids, pH 2.0-4.0, further milled in a blender, and mixed with the weighting material-titanium or stainless steel particles 10-40 μm in diameter. This suspension is then formed into droplets of the desired size and frozen, usually in liquid nitrogen; the pore size is influenced by both pH and freezing rate. Additional control over pore size can be achieved by annealing the particles at -10°C to -20°C for at least four hours, after the initial freezing in liquid nitrogen. The particles are freeze-dried and crosslinked by a combination of dry heat and chemical treatment, ideally by carbodiiamide or succinimidyl active esters, then washed and sterilized. (Dean et al.) C. Biomat Informatrix porous microcarriers Basic processing of collagen-glucosaminoglycan materials begins with a thoroughly blended suspension of collagen, preferably from limed bovine hide in dilute acid at about 0.25-0.50%(w/v). A solution of glucosaminoglycan, usually chondroitin 6-sulfate, is slowly added, forming a co-precipitate. This suspension is then formed into sheets(wound dressings) or droplets(microcarriers) and frozen at a controlled rate to create ice crystals that generate the voids can vary from~1μm to ~800μm in diameter, with a void volume fraction of > 99%.
