ELISA
(= enzyme-linked immuno-sorbent assay)
A very sensitive technique for the detection of small amounts of protein or
other antigenic substance. The basis of the method is the binding of the
antigen by an antibody that is linked to the surface of a plate. Formation
of an immune complex is detected by use of peroxidase coupled to antibody,
the peroxidase being used to generate an amplifying colour reaction. Various
ways of carrying out the assay are possible: if the aim is to detect antibody
production from a myeloma clone, for example, then the antigen may be bound
to the plate, and the formation of the antibody/antigen complex may be
detected using peroxidase coupled to an anti-Ig antibody.
