: : 染後的病理變化,Cytopathic Effect(CPE).
: : (b)練習Hemadsorption test,觀察Hemadsorption現象.
: : 原理:
: : (a)cytopathic effact:病毒感染細胞後,會對細胞造成影響,並因而成為
: : 可用一般倍數顯微鏡可觀察到的病徵而判斷病毒的存在.這些影響小至細
: : 胞聚集,嚴重可能造成cytolysis.
: (b)homadsorption:在病毒感染細胞,並自細胞釋出前,可能因病毒產生的蛋白
: 鑲入被感染的細胞膜上,造成細胞膜上具有可吸附細胞的物質,因而造成細胞彼此
: 聚在一起的現象.此外,特定的病毒感染會造成特別種類的細胞發生此現象.hemadsorption
: test即利用這種現象來判斷病毒的存在及感染.
: 材料:(a)EO Cell
: IPNV L-15-2 medium+2%FBS PBS buffer
: Laminar flow Pipets 1ml,5ml,10ml
: Pipet add Incubator 18 c
: (b)BHK(baby hamster kidney) cell
: NDV(newcastle disease virus)infected cels
: 0.1%chicken RBC in saline
: Pipets 1ml,10ml,steriled
方法:
(a)
1.挑選一瓶EO Cell,將flask中的medium倒掉,以約3c.c.的PBS清洗細胞表面
,後將PBS倒乾淨.
2.加0.5c.c.IPNV,輕搖flask後在incubator 18 c靜置30分鐘.
3.再以PBS清洗之,倒乾淨後加入L-15-2 2%serum 5c.c.,再放回incubator培養.
4.隔一段時間後以顯微鏡觀察其CPE.
(b)
1.將裝BHK Cell之flask中之medium倒掉,以PBS洗BHK Cells,倒掉PBS.
2.加入0.5c.c.雞紅血球(0.5%),靜置30分鐘.(血球取用前先搖晃或輕彈
管壁,可減少血球沉澱情況)
3.以倒立顯微鏡觀察.
