標題[求救] transformation 完菌長不起來

我將plasmid transformation到yeast
以抗生素G418篩選
長出來後colony PCR check完 確實轉進去了
也批了yeast chromosome的DNA確定它是yeast
結果此colony點出來要另外放大卻一直長不起來
在YPD-G418 plate或液態medium都是
放了3天還長不起來
想請問大家也有遇過這樣的情況嗎?
這中間哪裡有出問題嗎?
